肉苁蓉中松果菊苷的制备及含量测定 【摘要】 目的:研究肉苁蓉中松果菊苷的制备方法。方法:采用微晶纤维素色谱与半制备高效液相色谱组合分离技术,分离纯化松果菊苷,采用薄层色谱法和高效液相色谱面积归一化法检测纯度,波谱法鉴定其结构,反相高效液相色谱法测定其含量。结果:制备的化合物鉴定为松果菊苷,纯度为:98.4%;含量测定在0.081 μg~0.81 μg呈良好线性关系,r = 0.9999,平均回收率为103.5%,RSD为2.60%,n = 6。结论: 该制备方法效率高,所得化合物纯度高,可用于大量制备;含量测定方法准确,简便,可靠,重复性好。【关键词】肉苁蓉;松果菊苷;微晶纤维素;半制备高效液相色谱法;反相高效液相色谱法;含量测定Preparation and Quantitative Analysis ofechinacoside Standard Sample in Cistanche 【Abstract】 Objective: To study apreparation and determination method of echinacoside in Herba Cistanches.Method:The echinacoside was separated and purified by semi-preparative highperformance liquid chromatograph and microcrystallinecellulosese chromatography. Its purity was determined by TLC and HPLC analysis andnormalization of peak areas. The structure of echinacoside was identified byspectrum, and the quantification was determined by RP-HPLC.Results:The compound was identified as echinacoside .The purity of the echinacoside was98.4%. There was agood linearity between 0.081 μg~0.81 μg (r = 0.9999).The averagerecovery was 103.5%(RSD = 2.60%,n = 6) . Conclusions: This preparation method isquick and convenient for producing the pure compounds from herbs with largecapacity quantitative. The method is accurate and simple with goodreproducibility, and it can control the quality of this herb and preparationeffectively.【Key words】Cistanche;echinacoside; microcrystalline cellulosese;semi-preparative high performance liquidchromatograph;RP-HPLC;   quantitative determination 肉苁蓉(Herba Cistanches)为列当科植物肉苁蓉Cistanche deserticola Y.C.Ma的干燥带鳞叶的肉质茎,近代药理学研究表明,肉苁蓉具有广泛的免疫调节、抗氧化、增强体力、抗疲劳等药理作用,在防治神经退行性疾病上具有较好的应用价值。肉苁蓉中的主要成分为苯乙醇苷类,松果菊苷在此类化合物中含量最大,因此我们选择肉苁蓉中的松果菊苷作为本品含量测定的指标成分。苯乙醇苷类是一类由苯乙基、咖啡酰基和若干个糖基组成的分子量较大的、结构彼此相似的水溶性成分, 常含有羟基、甲氧基等取代基,这类化合物极性相近,完全通过依靠极性分离的硅胶柱难于将其完全分开,且在硅胶上有很强的吸附,使得含量本来就极低的此类化合物损失较大,同时苯乙醇苷类化合物分子内都含有两个或两个以上的糖基,糖苷类化合物难于结晶,纯化较困难, 成为制约其科学研究及工业应用的瓶颈问题。微晶纤维素是天然纤维素经稀酸水解并经一系列处理后得到的葡萄糖的聚合体。由于微晶纤维素具有良好的物理和化学稳定性,对含糖基类的化合物吸附稳定,我们尝试以微晶纤维素为层析介质分离纯化苯乙醇苷类化合物,试图寻找一种分离该类化合物的新方法。本文现将肉苁蓉中松果菊苷对照品的制备方法和含量测定方法报道如下:1 仪器与试剂X-4型显微熔点仪(国产,温度计未校正);Waters2475半制备高效液相色谱仪,Angilent1100高效液相色谱仪,INOVA-400型核磁共振仪 (TMS为内标); HP 5973质谱仪 (电离条件为70 eV);KQ-250B型超声波清洗仪(昆山市超声仪器有限公司);D101大孔树脂(青岛海洋化工厂生产);微晶纤维素;聚酰胺薄层层析板;高效薄层层析板(GF254,青岛海洋化工厂生产);甲醇、乙睛为色谱醇,其他化学试剂均为分析醇,水为纯净水。肉苁蓉药材采自新疆,经贵阳中医学院陈德媛教授鉴定为列当科肉苁蓉属植物的干燥肉质茎。2. 方法与结果
